Development of In Situ Gelling Meloxicam-Human Serum Albumin Nanoparticle Formulation for Nose-to-Brain Application

چکیده

The aim of this study was to develop an intranasal in situ thermo-gelling meloxicam-human serum albumin (MEL-HSA) nanoparticulate formulation applying poloxamer 407 (P407), which can be administered liquid state into the nostril, and increase resistance against mucociliary clearance by sol-gel transition on nasal mucosa, as well improve drug absorption. Nanoparticle characterization showed that formulations containing 12–15% w/w P407 met requirements administration. Z-average (in range 180–304 nm), narrow polydispersity index (PdI, from 0.193 0.328), zeta potential (between ?9.4 ?7.0 mV) hypotonic osmolality (200–278 mOsmol/L) MEL-HSA nanoparticles predict enhanced absorption through mucosa. Based rheological, muco-adhesion, release permeability studies, 14% (MEL-HSA-P14%) considered optimized formulation, allows MEL blood–brain barrier-specific lipid fraction. Cell line studies no cell damage after 1-h treatment with MEL-HSA-P14% RPMI 2650 human endothelial cells’ moreover, permeation (four-fold) observed comparison pure MEL. Overall, promising for overcoming challenges delivery.

Prolonged Release Evaluation of an Injectable Anticancer Drug using Human Serum Albumin Nanoparticle

Human serum albumin nanoparticles (HSA-NPs) were synthesized using the modified desolvation method. Fourier transform infrared spectroscopy (FT-IR), electronic absorption spectroscopy (UV-Vis), Zeta Sizer as well as field emission scanning electron microscope (FE-SEM) of the sample confirmed the formation of HSA NPs with an average size of 68 nm. The obtained results shown that HSA-NPs was succ...

Human Serum Albumin-Based Nanoparticle-Mediated In Vitro Gene Delivery

The genetic treatment of neurodegenerative diseases still remains a challenging task since many approaches fail to deliver the therapeutic material in relevant concentrations into the brain. As viral vectors comprise the risk of immune and inflammatory responses, human serum albumin (HSA) nanoparticles were found to represent a safer and more convenient alternative. Their ability to cross the b...
